Part 2 Chemistry Webinar Series: Remote Access Robotic Controlled Compound Synthesis - Recap

On September 15th, Strateos hosted Part 2 of its Chemistry Webinar Series: Remote Access Robotic Controlled Compound Synthesis. In continued efforts to provide scientists with end-to-end technologies to accelerate their workflows, Strateos has expanded its synthetic chemistry solutions with the integration of PerkinElmer Informatics Signals™ electronic lab notebook (ELN). Ben Miles, Ph.D., Head of Product Management at Strateos was joined by Josh Bond, Director of Product Management at PerkinElmer Informatics to discuss this exciting collaboration.

OVERCOMING BOTTLENECKS IN TRADITIONAL WORKFLOWS

Conventional small molecule drug discovery workflows take upwards of 6 years to progress from target ID through to IND submission with key medicinal chemistry (MedChem) steps involved in lead generation. However, bottlenecks in traditional MedChem workflows like slow synthesis and testing cycles, non-standardized data, and lack of reproducibility remain a challenge.

Supporting early lead generation through the provision of remote access on-demand robotic automation modules with integrated tools and technologies on one platform, Strateos enables the rapid synthesis, purification, and screening of drug compounds. Software control and data standards built into the Strateos platform generates reproducible model quality data that can feed into novel AI/machine learning and design tools to accelerate the design-make-test-analyze cycle driving next generation drug discovery.

STRATEOS AND PERKINELMER COLLABORATION: THE BEST OF BOTH WORLDS

The combination of PerkinElmer Informatics Signals™ Notebook ELN with the Strateos Platform provides users with the benefit of the ELN functionality for experimental design and Strateos’ automated synthesis and testing modules, enabling rapid iterative rounds of compound synthesis and screening. This complementation also overcomes the fragmentation of traditional workflows (mixture of in-house and CRO operations) to streamline workflows, condense timelines, maximize accessibility, and increase data transparency.

Ben and Josh walk through a use-case example of how a user would design a synthetic reaction in the Signals ELN and submit it to Strateos. Stoichiometric parameters entered into the ELN required by the robotic modules along with reaction specifications are delivered to the Strateos console with click-button submission.

Final experimental configuration occurs on the Strateos interface where software prompts allow users to flexibly choose custom (provided by user to Strateos) or commercial material selected from the built-in searchable e-molecules catalog.

Strateos SmartLab Studio reaction modules move materials downstream from synthesis to solid phase extraction and structure verification with NMR and LCMS that can feed directly into assay studies to automatically generate multiparameter data on a compound of interest. Data and metadata (i.e., instrument settings) captured throughout the experiment can be analyzed with onboard analytics tools or pulled back into Signals ELN for further manipulation. Reproducibility, traceability, and continuity of data from design through to results is maintained.

The goal of automating and working towards closing the design-make-test-analyze loop facilitates the generation of clean, AI-actionable data that overcomes the bottlenecks in small molecule compound design and synthesis, and purification workflows.
